Col de Lautaret: fastest way through the mountains
You can choose the D1091, Grenoble - Briançon via the Col du Lautaret. A beautiful road through the mountains from Grenoble. It's not far, but the drive will take about 3 hours. Don't mistake it for this. This road can get stuck or closed in bad weather (snow and wind). In that case, consider driving via the Fréjus tunnel.

Frejus tunnel: fewer mountain roads, but 30 min longer and tolls
You can go via the Fréjus tunnel on the A43. Then you pass Italy and the Col de Montgènevre. This road is no faster than via the Col du Lautaret, but you're on the motorway longer and less on mountain roads. This tunnel costs for a passenger car €44 single way, €55 for a return (note! returns are only valid for 7 days).

Route Napoléon over Gap: no col over, 45 min longer
You can also take the Route Napoléon: from Lyon, take the A43/A48 to Grenoble, then towards Gap, then Embrun and Puy-St-Vincent. This route takes about 45 min longer. With this route, you don't have to cross a real col.
